Bullet Seating Depth 45 colt

Hey guys, I'm going to start reloading 45 colt soon and got a lee mold for cast
WFN GC 300 grain bullets. the problem is that it has two crimp grooves, a helpful tool for any needed variations in bullet seating depth. However, I'm new to the reloading world and would appreciate any information regarding how deep I should seat my bullets. I'm not sure if air space in the shell effects pressure, or if it is just a way to compact certain powders or to adjust for varying OAL adjustmens. Will be making loads w/ aprox. 16-22 grains of powder. Thanks
