I recently came into a Cooper Firearms Model22 in .22-250 Ackley improved with a Leupold VXIII 6.5x-20x varmint reticle. I keep hearing that I need to fire form brass which I intend on doing. My question is am I still able to shoot .22-250 Rem or am I bound to fire forming brass?

I was thinking on the lines of .223 and 5.56mm being interchangeable to certain degrees, i.e. .223 in a 5.56 but not vice versa.

Am I off, or is it ok. Reason I ask is I was wanting to shoot coyotes while fire forming brass. It has a custom 1-9 twist so I can run the heavier rounds for med game animals and such, but I don't want to derp my chamber.

Any thoughts?

That is the whole point of the AI series, you can shoot factory loads or full charge handloads in them with only a slight loss in velocity, little or no loss in accuracy, and get fireformed brass to reload.
