Given only one choice, It would be the .357 magnum.

To open up the "ONLY ONE" a bit, my choice would be a 12 gauge shotgun with loads from bird shot to slugs and buckshot in between.

Caliber - .44. Cartridge .44 Special. BUT, though the OP instructed that platform (use, etc) was not to be considered, it almost has to be. While, cartridge-wise the .44 Sp will do everything I ask--or can conceive of asking--a .357 to do, the latter would be my choice if a carry handgun - 1-2 more in the chamber, generally lighter gun, off-the-shelf ammo availability, etc. (Hard times, emergency/Post Sandy/Katrina, etc).

Back to the OP,...when following instructions that such "operational" characteristicss are not under discussion, just strictly caliber (or cartridge which is probably what he meant) chracteristics, I'm back to .44/.44 Sp. - hands down. Same "effective effectiveness" as the .357, less sturm-und- drang, chance you might get your hearing back if shot indoors or even caught "having" to shoot outdoors without haring protection. Decent SD-thru-game hunting energy. Larger hole. Better range (and loading variety) than the otherwise very similar .45 ACP. What's not to like?--again just talking cartridge, not gun. Strictly caliber discussion (vs gun) opens this up to .44 Mag. As a previous poster said, load it up or down, most versatile cartridge out there, especially when availability is taken into account. I'll grant the .45 (Colt) boys will have a point arguing at least parity if you reload.

I'm definitely taking the .41 Short Rimfire.

Why? Everything else was taken, I don't want to have to compete at the counter for my ammo and also because the bullet moves SO SLOWLY that I can simply pick up all my slugs after shooting and DING - instant source of range lead without all the work to mine a berm.

While the 357 gets my vote I think a 375 magnum could make the most of the medium frame single actions.

I can not believe that no one has said 10mm. You can load them light for plinking or nuclear to kick some major tail.

True, and while it would be my second choice, my first will always be .45 ACP, and my ultimate handgun exists already, the 1911. Not made in every caliber, but a bunch.

I would have also picked my beloved 10mm but I looked at the whole picture and even though I reload, I took into consideration of cost and availability of ammo.

41 magnum I could load it light with 175gr silvertips or hot rod it with a 260gr hardcast.

Depends on it's purpose.

If I have to choose one to do it all, I'd agree with 10mm.

If hunting Kodiak bear, .500 Smith; if hunting squirrel, .22 short; self defense, .45 ACP. Everything else falls in between.

It would depend on my circumstances. With my current circumstances, probably a 9mm or even a .380. For other circumstances in which I've lived, it might be a .357 or a .45 ACP. But both my father and my father-in-law somehow managed with only a single .22 rimfire each and both lived in the country. So I don't really like to be dogmatic about such things.

At the moment, I actually own two 9mm automatics, so while I may not be dogmatic, I am at least decisive. Sometimes.

Best all-around round? I'd say 357.

If the circumstance for having one calibur only was armageddon and a complete breakdown of civilization, I think I'd grab a 9mm. It seems that everybody and his uncle has one. If I had to search a dead body for ammo, it's not likely I'd find 357's.
Of course, with my luck, everywhere I searched, I'd only find 22WRF's.

.357mag

Probably a revolver chambered in .357 mag. Ammo is plentiful, reasonably priced and I can also can use 38spl. in the gun. I can also use the ammo in a lever action Winchester rifle.

Less recoil than a 9mm, hits with almost as much force as a .357 magnum.

The terminal ballistics of the Hornady V-Max round is utterly devastating.

2 or 4 legged threat, It would do the job.

If that charging bear doesn't change its mind about charging pretty quickly, its brain is going to be turned to mush by the tremendous shock imparted by the bullets.
