Bump the wedge out from the side opposite the screw, no need to remove the screw.With the screw in place it won't come completely out but once it's cleared the arbor slot put the revolver on half cock. Turn the cylinder till it's lined up with the ram between chambers. Lower the ram and use it to pry the barrel off. Now the cylinder will slip off the arbor. When you reinstall the barrel you might have to use a piece of wood to bump it back down on the pins. The wedge should be flush on the side opposite the screw. Drive it in too far and it will bind the cylinder.
