Whatever you already have reloading dies for or whatever matches the ammo your six-shooters use. There's nothing like competing in cowboy action shooting and having to carry different ammo for each gun.

If you don't reload and you want to hunt deer with the gun, I would choose the .44 magnum. Factory loads are a lot more powerful than the .45 Colt and when you feel like just plinking, .44 special ammo should work. The only difference is the case is about 1/10 inch shorter.
