I was considering a Ruger Single Six as a purchase option and was wondering if anyone might be kind enough to post some side by side comparison photos next to a Vaquero so that I can get an idea about the actual size.

Top is a short grip Single Six in .32 H&R mag.
Bottom is an original Vaquero in .45 Colt.
Both have 4 5/8" barrels.

As he said, the single six shown is sporting the short grip many (but by far not all as many will eroneously have you believe) .32s came in. The Single Six's "regular" grip in the .22s as well as .32 is the same as the large frame Vaquero's, which is--in turn--the same size grip and frame as modern Blackhawks, except the 50th Ann. .357 (same as the mid-frame New Vaqueros' grip and frame).
The Single Six is virtually identical in overall size as the shiny toy six shooters many of us baby-boomers grew up with (except the grip size I would guess!) --my SS fitting into a holster (I still use!) my father made for me in '59--like a glove, including his original wet moulding around the cylinder and ejector rod!
You didn't mention the New Vaquero but there's another choice...about 1/4-1/3 the way in size between the SS and original Vaquero shown (at about 5', the NV looks about the same as the Vaquero). The 50th Ann..44 Blackhawk is a sort of hybrid: has the same gripframe as the smaller NV and 50th .357 but mated to the standard/larger "main" (cylinder) frame as all other Blackhawks and original Vaqueros. The latter is, functionally and structurally, "simply" a smoothed, fixed sighted Blackhawk.
